ABSTRACT

Over the past 20 years, amino acid analysis methods based on precolumn derivatization procedures have become a popular standard practice in many biochemistry laboratories as practical alternatives to the traditional older postcolumn derivatization techniques based on ninhydrin detection following the ion-exchange separation of the underivatized amino acids. The major reasons for this enthusiastic endorsement include greater sensitivity, faster analyses, and the ability to use less expensive, more flexible high-performance liquid chromatography (HPLC) instrumentation rather than dedicated amino acid analyzers.
