ABSTRACT

Protoplasts from adult leaves of lettuce were electroporated in osmotically adjusted and buffered solutions containing plasmid DNA of either pCAMV CAT or pABDl. Transient CAT activity measurements and selection for kanamycin resistance showed that expression of foreign genes and stable transformation were achieved. Analysis of the progeny of regenerated transformed lettuce showed that resistance to kanamycin is inherited as a dominant Mendelian trait. Integration of foreign DNA into genomic lettuce DNA of resistant plants was demonstrated by Southern blot hybridizations. Thus lettuce protoplasts are amenable to direct transformation under the same general conditions as those employed for tobacco protoplasts. Direct transformation provides new opportunities in some specific strategies for lettuce breeding. More... »
