ABSTRACT

Embryonic stem cells: The ground state for self-renewalEmbryonic stem (ES) cell self-renewal has generally been viewed as being dependent on the stimulation of various transcriptional pathways by exogenous factors in the culture media. A new study of mouse ES cells in a variety of cell culture condtions shows that, in fact, ES cell self-renewal does not rely on extrinsic instruction, and can be enabled by the elimination of factors that induce differentiation, and by inhibiting glycogen synthase kinase 3. This suggests that the ground state of the ES cell in the absence of extrinsic instruction is self-renewal — a display of self sufficiency that is more like that of a unicellular organism than the interdependence we expect of regular metazoan cells.
