ABSTRACT

THE specific interaction of many components is necessary for the assembly and function of flagella in Escherichia coli. Twenty genes have been shown to participate1–3 and at least eleven polypeptides are associated with the structure of the flagellar organelle4. The synthesis and assembly of these components and of flagellin, the major flagellar subunit protein, is tightly coupled and regulated5–7. It has not been possible to analyse these regulatory mechanisms in great detail because: (1) the tight coupling between events usually leads either to the synthesis of a complete organelle, or to no detectable flagellar structure, and (2) most of the flagellar components are present at low levels and their synthesis is difficult to measure. To analyse the hierarchy of control and regulation in this system we have prepared a series of hybrid transducing phages that carry different combinations of flagellar genes. We describe here one of these phages and show that it can be used to study the expression of flagellar gene activities. More... »
