AUTHORS ABSTRACTBACTERIAL lipopolysaccharides (LPS) stimulate DNA synthesis specifically in B (thymus-independent) lymphocytes1–4, increase immunoglobulin synthesis and secretion in B cells5, and stimulate the immune response of antibody production to various antigens in the apparent absence of T (thymus-dependent) lymphocytes4,6–8. Recently a mouse B lymphosarcoma, PU-5-1, has been described which has surface immunoglobulin9, and produces a small amount of immunoglobulin (R. Asofsky, unpublished). Cells of this tumour were adapted to culture and tested for the LPS stimulation of immunoglobulin secretion. Surprisingly, growth of the tumour cells was inhibited by LPS, although there was no effect on mouse myelomas or T lymphomas. Inhibition also occurred with an Abelson leukaemia virus induced tumour believed to represent a B lymphoma10 or immature myelomonocytic neoplasm11. No effect was observed on the growth rate of a number of human B cell lines tested. LPS is not cytotoxic for the tumour cells: growth resumes when it is removed. Incorporation of precursors into DNA is inhibited before RNA and protein synthesis is affected.
