ABSTRACT

Apoptose ist ein essentieller Prozess während der Embryonalentwicklung und der Aufrechterhaltung der Zellhomöostase in multizellulären Organismen. Der extrinsische Signalweg der Apoptose wird durch die Stimulation von Todesrezeptoren wie CD95 (APO-1/Fas) ausgelöst und führt zur Bildung des Tod-induzierenden Signalkomplexes DISC („death-inducing signaling complex“) an der Zellmembran. Am DISC wird Caspase-8 aktiviert und initiiert das apoptotische Programm. Defekte in der CD95-Signalleitung führen zur humanen lymphoproliferativen Autoimmunerkrankung ALPS („autoimmune lymphoproliferative syndrome“) und verdeutlichen die wichtige Rolle von CD95 in der Lymphozytenhomöostase. Agonisten gegen andere Todesrezeptoren, wie beispielsweise TRAIL-R („tumor necrosis factor-related apoptosis- inducing ligand receptor“), haben in der Behandlung von Tumoren bereits in klinischen Studien Wirkung gezeigt. CD95-spezifische Therapien werden ebenfalls entwickelt. Bispezifische Liganden, die sowohl CD95 als auch Tumorstromaproteine binden, könnten neue therapeutische Möglichkeiten eröffnen. In den letzten Jahren mehren sich die Hinweise darauf, dass CD95 neben der Apoptoseinduktion auch nichtapoptotische Signalwege aktivieren kann. Zukünftig wird es entscheidend sein, die differentielle Signalleitung von CD95 genauer zu untersuchen und diese Erkenntnisse bei der Entwicklung von therapeutischen Anwendungen einzusetzen.
