ABSTRACT

The fluorescent dye 1-dimethylaminonaphthalene-5-sulphonyl chloride was synthetized and the conditions of its conjugation with proteins were examined. Whereas the concentration of protein in and the ionic strength of the medium had no appreciable influence upon the efficiency of binding the reverse was true of pH. Throughout the experiments one kind of immune γ-globulin was used for conjugation, the immunological properties of which did not change upon chemical interaction with dansylchloride. 3–9 molecules of the fluorescent marking substance were bound to one molecule of the γ-globulin depending upon the conditions used. Prolonged standing at +4°C of the dye-protein complex resulted in a slight dissociation which had no marked influence on the staining qualities of the conjugate. More... »
