ABSTRACT

A new technique for doubling the chromosome number of potato clones was used to treat 54 diploid (2n−24) genotypes selected for desirable tuber characteristics and yield. Scions of the clonal selections were grafted onto tomato stocks. After the scions had grown 25–30 cm, all visible growing points were removed. The nodal, sub-axillary meristems present at the bases of the pseudostipules were treated with colchicine solution and allowed 8 weeks to regenerate new growth. The shoots produced by treated sub-axillary meristems were detached, rooted, and allowed to tuberize. Plants derived from the tubers were screened for tetraploids by 1, root-tip chromosome counts, 2. seed set following self-pollinations and cross-pollinations with diploid and tetraploid pollen parents, and 3. stomatal measurements. Chromosome-doubled genotypes of 33 of the clones (61% success) were identified and preserved. The technique appears to be the most efficient method of clonal doubling reported thus far. More... »
