ABSTRACT

In short Purkinje fibers tetrodotoxin (TTX) is only effective in concentrations >10−6g/ml. At 10−5g/ml TTX the rate of rise of the action potential is reduced to about 10%. The plateau phase is shortened, while the pacemaker potential is not affected. Even after long application of 2 · 10−5g/ml the effects of TTX are fully reversible by washing.The negative excitatory (inward) sodium current flowing during voltage changes enforced by a voltage clamp is nearly abolished in the presence of TTX. In the sodium-free solution a positive (outward) sodium current is observed which is reduced in presence of TTX. In the steady state after a voltage step or during very slow de- and repolarization at constant speed TTX does not have an appreciable effect on the currents measured in Tyrode's solution.The difference of the currents measured in Tyrode's in presence and absence of TTX is interpreted as sodium current. Following a superthreshold voltage step sodium current declines to about 1% within 10 ms, after further 140 ms to 0.1%.The increase of positive membrane current on removal of extracellular sodium cannot be attributed to lack of negative sodium current. Under this condition a negative current decreases which is not carried by sodium ions, in addition to possible effects on the potassium current. More... »
