ABSTRACT

Solutions of polystyrene and dextran (molecular weight approx. 2·106) were investigated by electron-microscopy in order to check the hypothesis which states that high polymer molecules are composed of subunits with a characteristic size of 30–70 Å. On well focused micrographs of negative-stained specimen it was neither possible to observe individual macromolecules nor could a clear distinction between specimen containing the polymer and blanks be established. Only defocused pictures showed structures which were identical with those which have frequently been used in the literature to prove the subunit-hypothesis. However, since these phase structures can be observed on specimen containing the high polymer molecules as well as on empty layers of negative-stain, the underfocused pictures by no means verify the subunit-hypothesis.The relation between the degree of defocusing and the spatial frequencies which are transfered with optimum contrast were extended to an underfocus of 3 μ.
