ABSTRACT

Summary1. In isolated frog atria the effect of 2,4-dinitrophenol (0.2 and 2 mMol/l) and other inhibitors on the K and Na fluxes, the membrane and action potential, and the intracellular level of high-energy phosphates was studied. The external K and Na concentration was varied within a wide range.2. Dinitrophenol (DNP) decreases K influx by about 30% but increases K efflux by 50 to 270%. Ouabain, sodium azide, and Li (replacing extracellular Na) lead to similar effects on the K exchange. Sodium iodoacetate has no marked effect.3. DNP decreases Na efflux by about 25%. Na influx is increase or decreased depending upon the composition of the external solution especially with respect to the Ca concentration.4. DNP produces a slow depolarization of membrane potential (after an initial phase of transient hyperpolarization). Simultaneously a progressive shortening of action potential occurs. After some minutes the excitability is abolished. The shortening of action potential corresponds to the increase of K efflux.5. The exposure of the preparations to DNP over some minutes causes a complete break-down of creatine phosphate, a decrease of ATP by about a fourth of its original level and a markedly increase of AMP and Pi. The quotient high-energy to low-energy phosphate was reduced to about 1/8 by DNP.6. When the inulin, mannitol, and sulphate uptake of the preparations were measured, there was no indication that the cell membrane had been damaged by DNP.7. It is concluded that the DNP-induced decrease of K influx and Na efflux is based on an inhibition of active K and Na transport. The reason for the increase of K efflux is not clear; the two possibilities of an augmented passive K permeability and an inversion of active transport were considered.8. A quantitative comparison of K inward and Na outward transport over the range of external solutions suggests a variable rather than a tight coupling and therefore an electrogenic nature of the pumps.9. An analysis of K transport based on a model of a circulating carrier system (Haas, 1967) results in a further hint to an electrogenic K pump. More... »
