ABSTRACT

Am Präparat der isolierten, zellfrei perfundierten Rattenniere wurde die Wirkung von Senkungen der Na+-Konzentration des Perfusions-mediums und die des Zusatzes von g-Strophanthin (Ouabain) zum Perfusions-medium auf die Glucoseresorption untersucht. 1. Schrittweise Senkung der Na+-Konzentration des Mediums bewirkt von Na+-Konzentrationen <50 mMol/l an abwärts eine fortschreitende Abnahme der Glucoseresorption bei konstantem tubulären Angebot. Rund 25% des Angebotes werden selbst in praktisch Na+-frei perfundierten Nieren noch resorbiert. 2. Zusatz von Strophanthin (0,5 mg/ml) zum Perfusionsmedium führt zu einer Senkung der Netto-Na+-Resorption um 40%. 3. Die gleiche Strophanthinkonzentration erniedrigt die Netto-Glucoseresorption um etwa 30%. Zur Erklärung der Befunde wird die Na+-Abhängigkeit einer Na+-K+-aktivierten, glykosidhemmbaren ATPase diskutiert, welche die Energiefreisetzung sowohl für den aktiven Na+- als auch für den aktiven Glucosetransport steuert.     46 schema:description Am Präparat der isolierten, zellfrei perfundierten Rattenniere wurde die Wirkung von Senkungen der Na+-Konzentration des Perfusions-mediums und die des Zusatzes von g-Strophanthin (Ouabain) zum Perfusions-medium auf die Glucoseresorption untersucht. 1. Schrittweise Senkung der Na+-Konzentration des Mediums bewirkt von Na+-Konzentrationen <50 mMol/l an abwärts eine fortschreitende Abnahme der Glucoseresorption bei konstantem tubulären Angebot. Rund 25% des Angebotes werden selbst in praktisch Na+-frei perfundierten Nieren noch resorbiert. 2. Zusatz von Strophanthin (0,5 mg/ml) zum Perfusionsmedium führt zu einer Senkung der Netto-Na+-Resorption um 40%. 3. Die gleiche Strophanthinkonzentration erniedrigt die Netto-Glucoseresorption um etwa 30%. Zur Erklärung der Befunde wird die Na+-Abhängigkeit einer Na+-K+-aktivierten, glykosidhemmbaren ATPase diskutiert, welche die Energiefreisetzung sowohl für den aktiven Na+- als auch für den aktiven Glucosetransport steuert.
