ABSTRACTCrude extracts from λ-lysogens treated with mitomycin C were prepared, and immunity repressor levels in the extracts were assayed by the binding activity specific to λ DNA immunity region. It has been shown that while the repressor levels in the extracts from C600(λ+) are reduced after mitomycin C treatment, the levels in C600recA(λ+) and C600 C72(λ+) which have defects in lifting the immunity are not affected by the treatment. The repressor levels in the extracts prepared from C600 T44(λ+) after temperature shift up, whose prophage is inducible at high temperature, are also reduced. From the study of chloramphenicol effect, it was indicated that de novo protein synthesis is required for the inactivation of the repressor in C600(λ+) by mitomycin C, but not in T44(λ+) by high temperature.
