ABSTRACT

Using the system described in the preceding paper (Iida et al., 1970), we have shown that lac repressor fractions purified one hundred to several hundred-fold repress the transcription of the lac operon in vitro and that an inducer IPTG1 inhibits the repression. The amount of lac messenger RNA produced in the case of the most powerful repression is calculated to be 1/10 of that in the presence of the inducer. In contrast, when the template DNA carries an operator constitutive (oc) mutation, little or no repression was observed. An efficient repression requires besides a purified repressor fraction, the presence of a cellular ribosomal fraction and an adequate salt concentration. More... »
