ABSTRACT

We have developed a means to investigate individual subcellular organelles, using integrated chip electrophoresis (ICE), optical tweezers, and laser induced fluorescence (LIF). Using this methodology, we manipulate and lyse individual organelles, followed by derivitization, separation, and detection of organelle contents. We are currently investigating the secretory granules of mast cells. Our data will be the first of its kind for mast cells; we hope to gain insight into the mechanisms of various mast cell functions, such as antigen-driven exocytosis or infection-driven phenotypic change. In addition to mast cells, we anticipate that this method will be useful for any biological or environmental application that would benefit from probing the character and content of individual microparticles.
