ABSTRACT

Every type of plant tissue culture is an evoluting system where cell divisions, cell differentiation and development (organogenesis) processes take place during cycles on successive renewed, same or different, culture media. From the beginning of subculture to the end of the cycle, the “explants” are submitted simultaneously and/or successively to several stresses: injury from dissection, abnormal mineral nutrition and hormonal balance, high relative humidity, gas confinement,… Attempt to characterize a culture type of by a redox potential is therefore a nonsense. The present study -concerning the special case of hyperhydricity affecting shoots under multiplication — therefore aims at investigating the redox capacities or the redox signalling systems used by such shoots to survive. The classical redox couples AA/DHA, GSH/GSSG and NAD(P)H/NAD(P), together with corresponding enzymes activities of the Asada-Halliwell, glycolytic and pentose-phosphate pathways respectively, soluble reducing substances and several markers of stress, have been investigated comparatively in shoots kept normal close to hyperhydrating ones. The study has evidenced indirectly the participation of a fourth redox couple formed by ferredoxin and thioredoxin. The present study -concerning the special case of hyperhydricity affecting shoots under multiplication \u2014 therefore aims at investigating the redox capacities or the redox signalling systems used by such shoots to survive. The classical redox couples AA/DHA, GSH/GSSG and NAD(P)H/NAD(P), together with corresponding enzymes activities of the Asada-Halliwell, glycolytic and pentose-phosphate pathways respectively, soluble reducing substances and several markers of stress, have been investigated comparatively in shoots kept normal close to hyperhydrating ones. The study has evidenced indirectly the participation of a fourth redox couple formed by ferredoxin and thioredoxin. There is evidence of occurrence of circadian rhythmicity in the participation of such redox systems.",
