ABSTRACT

The glycosylation pattern of Erythropoietin (EPO), produced by recombinant CHO cell, was studied using rapid and simple method of ‘Lectin-blotting’. We used three different lectins in this experiment; MAA (Maackia amurensis agglutinine), RCA (Ricinus communis agglutinine), DSA (Datura stramonium agglutinine) which binds to terminal sialic acid, galactose and N-acetyllactosamine chain respectively. The effect of ammonium ion on glycosylation of EPO was examined since it accumulates in the medium as a byproduct mainly resulted from the glutamine metabolism. Consequently, treatment of ammonium chloride from 8mM to above substantially inhibited the sialylation of terminal galactose residue. More... »
