ABSTRACTThe liver is a key organ for the metabolism of drugs and endogenous substrates. When the liver sustains serious injury, many different parameters are affected, such as blood clotting, glucose metabolism, drug metabolism and brain function. To help maintain these life-sustaining functions, the liver has a remarkable capacity to regenerate following injury or loss of tissue mass. It has long been hypothesized that the liver changes its functional mass in response to changing metabolic demands. The regenerative capacity of the liver has been recognized and studied for decades (Bucher, 1963; Alison, 1986; Michalopoulos, 1990; Fausto, 1991), but only recently have we been able to gain more insight into the molecular basis for this process. Several growth factors have been identified which modulate hepatocyte proliferation. Using differential cloning techniques, a number of genes have been isolated that are rapidly activated after stimulation of hepatic regeneration (Nathans et al., 1988; Mohn et al., 1990, 1991; Haber et al., 1993; Weir et al., 1994). At present, we lack an understanding of the precise role of growth factors and ‘regeneration response’ genes in the regulation of liver regeneration, but pieces of this puzzle are rapidly being collected. This chapter focuses on factors that inhibit growth and/or stimulate programmed cell death of hepatocytes. Growth inhibition is an extremely important component of the regenerative response because once regeneration is initiated, hepatocyte proliferation must be turned off in order to prevent uncontrolled liver growth. "description": "The liver is a key organ for the metabolism of drugs and endogenous substrates. When the liver sustains serious injury, many different parameters are affected, such as blood clotting, glucose metabolism, drug metabolism and brain function. To help maintain these life-sustaining functions, the liver has a remarkable capacity to regenerate following injury or loss of tissue mass. It has long been hypothesized that the liver changes its functional mass in response to changing metabolic demands. The regenerative capacity of the liver has been recognized and studied for decades (Bucher, 1963; Alison, 1986; Michalopoulos, 1990; Fausto, 1991), but only recently have we been able to gain more insight into the molecular basis for this process. Several growth factors have been identified which modulate hepatocyte proliferation. Using differential cloning techniques, a number of genes have been isolated that are rapidly activated after stimulation of hepatic regeneration (Nathans et al., 1988; Mohn et al., 1990, 1991; Haber et al., 1993; Weir et al., 1994). At present, we lack an understanding of the precise role of growth factors and \u2018regeneration response\u2019 genes in the regulation of liver regeneration, but pieces of this puzzle are rapidly being collected. This chapter focuses on factors that inhibit growth and/or stimulate programmed cell death of hepatocytes. Growth inhibition is an extremely important component of the regenerative response because once regeneration is initiated, hepatocyte proliferation must be turned off in order to prevent uncontrolled liver growth.",
