AUTHORS ABSTRACTIn order to develop an efficient process for the production of recombinant proteins with the baculovirus expression system, various factors concerning the culture conditions of Sf-21 - a cell line derived from the “Fall Armyworm” Spodoptera fnigiperda were studied. These insect cells are very common in insect cell culture and were used as host cells for the infection with recombinant Autographa californica Nuclear Polyhedrosis Virus. To optimize the growth of the Sf-21 cells, four different media supplemented with 5% fetal calf serum (FCS) were tested in 125 ml spinner flasks. The highest cell density was achieved in IPL-41 modified with 2.5 g/l tryptose phosphate broth + 5% FCS. This medium was used for further continuous and batch experiments of Sf-21 cells in a perfused 1.4 l bioreactor. In these experiments the exponential growth phase could be maintained for more than 7 d and a maximal cell density of 2•107 viable cells/ml was achieved.
