ABSTRACT

Competition for iron plays an important role in bacterial virulence and animal host defense. Several attempts have been made to determine the role of microbial siderophores in plant infection, but only recently it has been clarified. In Erwinia chrysanthemi 3937, a plant pathogenic bacterium responsible for soft-rot diseases, Enard et al. (1988) have shown the relevance of an iron assimilation system mediated by the siderophore chrysobactin, in the virulence of this bacterium in saintpaulia plants. The level of iron in the intercellular fluid of plant leaves proved to be low enough to restrict the growth of iron uptake mutants during early stages of infection. To investigate the problem of iron acquisition in this pathogenic interaction, we undertook a molecular study of the chrysobactin system with a special emphasis on its regulation by iron. In addition, iron was found to modulate the expression of several pectinase-encoding genes of major importance for the virulence. We identified the presence of chrysobactin as well as the induction of the chrysobactin operon in infected tissues. Competition for nutritional iron was further studied through a plant-bacterial system from which drastic changes were shown to occur in iron metabolism of infected cultured soybean cells. More... »
