ABSTRACT

The majority of mitochondrial proteins are encoded by nuclear genes and are thus synthesized on cytosolic ribosomes as precursors containing an amino terminal extention (presequence) which is involved in targeting the precursor to the organelle. In vitro targeting systems have been used largely in Saccharomyces cerevisiae and Neurospora crassa [1,2] for unraveling the precursor amino acid structure and the protein machinery implicated in the targeting process. Although in plants in vitro targeting to chloroplasts has been used for a long time [3], a similar mitochondrial system has been developed much more recently, presumably because of the lack of cloned precursor sequences.
