ABSTRACT

The origins of immunocytochemistry can be traced back to the development of techniques for the labelling of antibodies with tags visible under the microscope. A close operational link between biochemistry and cytology was thus established, consisting of a classical, three-step experimental design: biochemical purification of individual molecular species (antigens) from tissues; production of antisera; identification of the molecular species in situ, exploiting an antigen-antibody reaction. In this procedure the specificity of the cellular staining is clearly based on the specificity of the antisera, which is in turn determined essentially by the purity of the antigen preparation.
