ABSTRACTSynthetic peptides (SP) based on the amino terminal sequence of a 3.8kD streptococcal antigen have been produced and used in topical oral immunisation of the gingivo-mucosal epithelium of macaque monkeys. Linear or cyclised SP11 and random SP11 induced negligible or very low antibody levels and did not prevent colonisation of the teeth by S. mutans. Immunisation with SP17, SP21 and SP35 induced significant anti-SP as well as anti-native streptococcal antibodies in the gingival fluid and saliva, and either prevented or greatly reduced, colonisation by S. mutans. The results suggest that local immunisation with SP derived from the sequences of a S. mutans antigen may induce a dual gingival IgG and salivary IgA response against SP and native SA, which may be involved in preventing colonisation of S. mutans and the onset of dental caries.
