ABSTRACT

The adsorption, in a liquid fluidized bed, of Bovine Serum Albumin, (BSA), onto an ion exchange adsorbent, consisting of quarternary (Q) amino groups, bound to an agarose matrix, (Sepharose®, Pharmacia LKB Biotechnology, Uppsala, Sweden), has been studied. The solid hydrodynamics have been examined with bed expansion tests. The equilibrium adsorption characteristics of the system have been assessed by the measurement of equilibrium adsorption isotherms which fitted well to the Langmuir equation. The rate of adsorption of BSA onto Q-Sepharose has been studied in a batch stirred tank, a fixed bed and a fluidized bed system. The data from the fluidized bed system has been used to assess the validity of a theoretical model adapted from one that predicts the performance of the adsorption phase in fixed bed systems.
