ABSTRACT

A system has been developed for the stable transformation of protoplasts of sugarbeet (Beta vulgaris) using electroporation-mediated direct gene transfer. Conditions for the reversible permeabi1ization of protoplasts, and the subsequent uptake of foreign DNA, were optimized by following the uptake of a) a hydrophilic dye, phenosafranine and b) the cat and gus reporter genes in transient expression studies. The frequencies of protoplast division and colony development were optimized, and stable integration of the aminoglycoside phosphotransferase gene, after electroporation, was demonstrated by the resistance of protoplast-derived colonies to kanamycin and by Southern blot analysis. Preliminary experiments were carried out to develop a shotgun cloning strategy for the isolation of selectable genes from plant tissue, using a direct gene transfer approach. More... »
