ABSTRACT

In vitro systems for Dactylis glomerata L. include the production of somatic embryos directly from mesophyll cells in cultured leaf explants and the complete development of embryos (to a germinable stage) in a single liquid medium. Regeneration has also been achieved from suspension-derived protoplasts and the plants grown to maturity. These systems offer attractive possibilities for gene transfer. The following experiments have been attempted: (1) abrasion of leaf surfaces and application of Agrobacterium vectors containing a kanamycin resistance gene (NPTII), (2) direct uptake of a DNA plasmid containing a hygromycin resistance gene (APHIV) by suspension-derived protoplasts, and (3) treatment of leaf segments and plated suspensions with microprojectiles coated with DNA containing both the NPTII and beta glucuronidase (GUS) genes. A few false positives were obtained with Agrobacterium and transient expression of the GUS gene was obtained with microprojectiles. The only stable transformants were produced through direct uptake of DNA by protoplasts.
