ABSTRACT

Electron transfer in photosynthetic reaction centers from Rhodobacter sphaeroides was investigated by time-resolved infrared spectroscopy using tunable lead salt diode lasers. In the spectral region between approximately 1690 and 1770 cm−1, transient µsec to msec signals could be characterized upon electron transfer from the primary quinone acceptor QA to he secondary acceptor QB. The dominant transient absorbance changes are three positive sig. s at 1725, 1735, and 1706 cm−1 and two negative signals at ca. 1716 and at 1698 cm−1. They are interpreted in terms of the C=O frequencies of aspartic or glutamic acid side chain residues affected by electron transfer. The major signal at 1725 cm−1 exhibits a fast kinetic component of t1/2 = 0.2 ms and a slow one oft t1/2 = 1 ms at pH 7. Analysis of its time course and amplitude under different conditions (pH, 1H/2H echange, mutations) allows its assignment to glutamic acid L212 near QB and indicates the uptake of a significant amount of a proton upon \documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$ {{Q}_{{{{B}^{ - }}}}} $$\end{document} formation.
