ABSTRACT

The seeds of legumes and most of the oilseed crops are “orthodox” and present no problem for long-term storage; however (1) “recalcitrant” species, in which seed is short-lived due to degeneration of the embryo, and where the germplasm cannot be conserved, require cryopreservation, (2) for the long-term preservation of elite and useful variants obtained by in vitro cell culture, innovative methods need to be evolved. Cryostorage of in vitro cultures has been recommended as one of the unconventional approaches for the long-term conservation of genetic stocks and for international exchange of germplasm (Bajaj 1976a, 1979a,b, 1983a, 1984a,b, 1985, 1986b). In this chapter, literature on the cryopreservation of tissue cultures of legumes and oilseed crops is reviewed, and prospects for this technology in the long-term conservation of germplasm are discussed.
