ABSTRACT

There is accumulating evidence that endogenous opioid peptides are involved in central and peripheral cardiovascular regulation. Yet, the precise nature of this involvement remains unclear. Acute stimulation of various opiate receptors has been shown to induce either decreases on increases in arterial blood pressure [1–4]. Centrally administered opiate antagonists such as naloxone produce a greater decrease in blood pressure in spontaneously hypertensive rats (SHR) than in normotensive Wistar Kyoto (WKY) rats [4, 5]. Intravenously applied naloxone, a preferential mu-receptor antagonist which also binds to delta- and kappa-receptors [6], was found to reduce blood pressure and heart rate in urethane-anesthetized SHR, but not in WKY rats [7]. Likewise, long-term administration of naloxone retards the age-dependent increase in blood pressure in young SHR, but not in WKY rats [8]. In some recent studies on SHR rats this retardation of the rise in blood pressure following chronic administration of naloxone could not be reproduced [9, 10].
