ABSTRACT

Fresh pollen of Nicotiana tabacum was cultured on Brewbaker and Kwack medium at pH 5,6 (control), 4.5, 4, 3.8, 3.6, 3.4 and 3.2, adjusted with H2SO4 Germination and pollen tube length of 100/150 grains was measured after 3/5 hrs of culturing. For TEM a standard glutaraldehyde/OsO4 technique and embedding Spurr’s resin was used. Microtubules were studied using monoclonal mouse anti-tubulin and rabbit anti-mouse antibody conjugated with fluorescein-isothiocyanate. Microfilaments were studied with labeling by phalloidin.
