ABSTRACT

Mutations are commonly used to enumerate and to understand the components in biological phenomena, including locomotion and behavior. In Paramecium, mutants have been isolated and studied, having in mind a genetic dissection of the behavioral mechanisms (Kung 1971a, b). To the genetic and behavioral analyses of the mutants was soon added an electric dimension, since the Paramecium membrane generates a Ca2+ action potential (Naitoh et al. 1972). The advent of the voltage clamp in this field greatly refined the electrical measurements (Naitoh and Eckert 1974; Oertel et al. 1977). Purification of the ciliary membrane was successful (Hansma and Kung 1975; Adoutte et al. 1980). However, the search for ion-channel proteins through the comparison of electrophoretic patterns of wild type and mutants was frustrated by physiological variations of membrane proteins (see Adoutte et al. 1983). In 1980, Hiwatashi and co-workers (Haga et al. 1982) reported success in restoring normal behavior in a mutant by microinjecting it with the cytoplasm of wild type. This functional assay has been used in a new search for the products of the cnrC gene (Haga et al. 1984 b) and the pntA gene (Hinrichsen et al. 1986).
