ABSTRACT

The Na+,K+-ATPase (NAKA) is an essential transmembrane enzyme complex that couples ATP hydrolysis with the transport of Na+ and K+ ions across the plasma cell membrane against their respective electrochemical gradients. The Na+/K+-, Ca2+ and H+/K+- active ion transport pumps of eukaryotic cells share significant similarities in tertiary structure, a modest degree of amino acid sequence identity, and similar enzymatic mechanisms that classify them as E1-E2 or P-type class of transport ATPases (12). The l00kDa a: 35kDa β complex constituting the Na,K-ATPase is encoded by at least three a isoform genes and two β subunit genes. These isoforms are characterized by high degrees of sequence homology and functional identity but with distinct tissue-specific, developmental and hormonal patterns of regulation. More... »
