ABSTRACT

Zur quantitativen Charakterisierung der synthetischen meso-Tetraphenylporphyrine TPPS1, TPPS2a, TPPS3 and TPPS4 in vitro an der Endothelzellime BKEz-7 (Kalbsaorta) wurden Zytotoxizität, intrazelluläre Akkumulation and Phototoxizität bestimmt. Die lipophilen Photosensibilisatoren TPPS1 and TPPS2a wiesen, im Vergleich zu TPPS3 and TPPS4, eine ausgeprägte Zytotoxizität auf. Die intrazelluläre Anreicherung war für alle vier Derivate, bei Inkubation mit der sensibilisatorspezifischen nicht-zytotoxischen Inkubationskonzentration, vergleichbar. Um eine photodynamische Inaktivierung auf 50% der Kontrolle zu erreichen, waren für TPPS1 and TPPS2a 10–20fach geringere Bestrahlungsdosen als für TPPS3 and TPPS4 erforderlich. Die ausgeprägten Unterschiede zwischen den lipophilen and hydrophilen Sensibilisatoren weisen auf eine unterschiedliche intrazelluläre Lokalisation and auf unterschiedliche Schädigungsorte in den Zellen hin.
