ABSTRACT

Secondary xylem vessel elements within the stems of grapevine (Vitis vinifera L.) mature over several weeks and stems maintain a large proportion of living vessel elements throughout most of the growing season. Only approximately one-third of secondary vessels within current-year growth are active at any single time. Once hydraulically active, the vessels of grapevine are highly susceptible to cavitation, consistent with the physiological response of most grapevine varieties to water stress. Nonconductive vessels may become blocked with either gels or tyloses. Large seasonal changes in both cavitation resistance and hydraulic conductivity occur in V. vinifera and may also be found along the length of a single stem, suggesting that the xylem may be able to dynamically respond throughout the season as growing conditions change. The high proportion of living vessels may be one of the reasons why interpretation of in situ imaging for grapevine has been difficult. Examination of the decline in the proportion of active vessels in response to water stress using traditional staining techniques confirms the high susceptibility of grapevine vessels to water stress and highlights the need for continued histological study of Vitis as new imaging technologies are developed. The high proportion of living vessels may be one of the reasons why interpretation of in situ imaging for grapevine has been difficult. Examination of the decline in the proportion of active vessels in response to water stress using traditional staining techniques confirms the high susceptibility of grapevine vessels to water stress and highlights the need for continued histological study of Vitis as new imaging technologies are developed.",
