ABSTRACTThis chapter mainly describes a cell-based treatment focused on a cultured corneal endothelial cell (CEC)-injection therapy, and it is devided into three parts; i.e., Background, Current State, and Future Directions. The background involves the successful culture of CECs in primates, animal-model experiments in which a cultured CEC sheet is used, and modification of the cultivation protocol and its use for CEC-injection therapy in an animal-model experiment. The current state involves cultured human CECs (HCECs) for clinical use, the clinical trials of cultured HCEC-injection therapy, the evaluation of the antigenecity of allogeneic cells and the fate of escaped cells from the eye post injection. As for the future directions, the ‘seeds and soil’ theory in the clinical setting, the combination therapy ewith ROCK-inhibitor eye drops, and the advancement of cell delivery and cell storage are all discussed.
