ABSTRACT

The fluorescence and phosphorescence of natural proteins arise from emission by the fluorogenic ring moieties of the aromatic amino acids. Such macromolecular luminescence may be visualized as corresponding to the intrinsic luminescence representing the summed contribution of the amino acid fluorogens, as perturbed by incorporation into a Polypeptide and by the influence of the secondary and tertiary structure of the protein. Accordingly, an understanding of the luminescence properties of tryptophan, tyrosine, and Phenylalanine is basic to an interpretation of the fluorescence and phosphorescence of the proteins. Such macromolecular luminescence may be visualized as corresponding to the intrinsic luminescence representing the summed contribution of the amino acid fluorogens, as perturbed by incorporation into a Polypeptide and by the influence of the secondary and tertiary structure of the protein. Accordingly, an understanding of the luminescence properties of tryptophan, tyrosine, and Phenylalanine is basic to an interpretation of the fluorescence and phosphorescence of the proteins.
