ABSTRACT

In the human red cell, ATP is an absolute requirement for the production of NAD. Thus, in pyruvate kinase deficient human erythrocytes NAD concentrations are significantly reduced and NAD synthesis is impaired to a degree that is dependent on ATP concentration of those cells (1). SCID is characterized by variable pancellular deficiency of adenosine deaminase (ADA). Red cells of SCID patients may have low ATP and high 2′dATP concentrations (2) but there are no reports on NAD or 2′dNAD content and synthesis in the se cells. D. virginiana is North America’s only native marsupial. Red cells of this species have low (1.5 nmoles/mg hg/h) ADA activity and high concentrations of 2′dATP resembling human SCID erythrocytes (3).
