ABSTRACT

The use of ancient DNA (aDNA) in the reconstruction of population origins and evolution is becoming increasingly common. However, contamination of ancient specimens and DNA extracts with modern DNA is more widespread than is generally acknowledged and remains a significant problem in aDNA analysis. Furthermore, studies of human aDNA are uniquely sensitive to contamination due to the continual presence of potential contamination sources and these studies require novel solutions for identification of contamination. Careful selection of polymorphic markers capable of discriminating between ancient DNA and probable DNA contaminants is critical. A promising new application of aDNA technology is the analysis of ancient human pathogens, which can be conducted with reduced risk of contamination. Fascinating questions to be addressed through ancient pathogen studies include the evolutionary history of pathogens and the migration patterns and evolutionary relationship of human populations.
