ABSTRACT

Molecular archaebacteriology is now about to enter a manipulative phase. Many laboratories have contributed to the development of methods for proper genetic analysis. Here we describe our efforts with Halobacteriurn volcanii. We have nearly completed a physical map of the genome of this species, have optimized procedures for its transformation by exogenous DNA, and developed shuttle vectors alternately selectable and maintainable in this species and Escherichia coli.
