ABSTRACT

The present level of understanding of the numerous biochemical cascades regulating processes of leukocyte-dependent inflammatory events has a great number of lacunae. Attempts to rationalize the many positive and negative feedback pathways by which the known proinflammatory molecular signals affect each other lead to complicated schematic diagrams which are of questionable relevance to any human inflammatory disease. However, a few concepts are worthy of discussion in order to consider the types of interactive effects that might occur between eicosanoids and cytokines. Whether these interactions are important in homeostasis and/or diseases may relate to the timing in the evolution of the process in terms of the activation state of the cellular target of the signalling molecule (e.g., the numbers of receptors expressed or the number of signal transduction molecules which are bioavailable for downstream events) as well as the concentrations of activating and inhibiting signal molecules in the microenvironment.
