ABSTRACTDark-grown angiosperms accumulate a well known chlorophyll precursor, protochlorophyllide (Pide), in the prolamellar body and in prothylakoids (Granick and Gassman, 1970; Brodersen, 1976; Lindsten et al., 1988). In vitro Pide occurs in two states: one is a non-photoreducible form noted P6 33 and the other one is a reducible form noted P657 (Sironval et al., 1968; Griffiths, 1980; Griffiths, 1981). P657 is usually described as a ternary complex between Pide, NADPH and NADPH protochlorophyllide oxidoreductase (PCR, EC 1.6.99.1) (Oliver and Griffiths, 1982; Griffiths et al., 1984; Ryberg and Sundqvist, 1988). When an etiolated leaf is illuminated by a short flash, PCR reduces P657 in a chlorophyllide form fluorescing at 688 nm (C688) as described by Sironval et al., (1968); Goedheer and Verhulsdonk (1970); Shoefs and Franck (1990). When the sample is maintained in the dark after an illumination, the maximum of chlorophyllide emission shows the well known rapid and Shibata shifts (Shibata, 1957; Sironval et al., 1967; Sironval et al., 1968).
