Intermittent hypoxia occurs in many breathing disorders including sleep apneas, asthma, chronic lung diseases and can have cardio-respiratory consequences including hypertension and altered ventilatory responses to hypoxia and hypercapnia.1,2 Despite its clinical significance, little is known about the molecular mechanisms associated with recurrent episodes of hypoxia. It is being increasingly recognized that induction of genes is one of the fundamental molecular mechanism that underlies physiological adaptations to chronic hypoxia.3 Recent studies have shown that the protooncogene c-fos is one such gene that is induced during hypoxia, and it regulates the expression of downstream genes (e.g., tyrosine hydroxylase) via activator protein-1 transcription factor.4 To begin to understand the molecular mechanisms associated with intermittent hypoxia, we asked whether recurrent episodes of hypoxia induces c-fos expression, if so, by what mechanisms. For this purpose, we exposed rat pheochromocytoma cells (PC 12) to intermittent hypoxia, and found that c fos is up-regulated by transcriptional activation which involves reactive oxygen species (ROS).
