ABSTRACT

The immunoregulatory activity of T cell subsets on human B cell differentiation has been amply studied in the pokeweed mitogen (PWM)-driven culture system (1,2). PWM-driven immunoglobulin (Ig) synthesis is strictly T cell-dependent and helper activity is only provided by T4+8−cells, whereas T4−8+ cells have suppressor activity (1,2). We demonstrated that interleukin-2 (IL-2) plays an important role in T cell-dependent Ig synthesis (3). It was found that PWM-induced IL-2 synthesis is an early and necessary event in PWM-driven Ig synthesis. This was confirmed by the finding that anti-Tac, an anti-IL-2 receptor antibody (4), fully abrogated PWM-driven Ig synthesis, and moreover, by the innate Iginducing capacity of IL-2 itself, dependent on T4+8− cells (3).
