ABSTRACT

Because of its relative simplicity and the wealth of information available on its membrane, the erythrocyte has served as a useful model for studies of the mechanisms of cellular and membrane fusion. Insight has been sought most often from scrutinizing intercellular fusion systems, while relatively little attention has been given to intracellular fusion and to what might be learned from evaluating conditions conducive to this type of fusion. Similarly, alterations in the protein portion of the membrane which might play a role in fusion have been given far more attention than the role which alterations in phospholipids, particularly their arrangement, might play in erythrocyte fusion. In this paper, we explore phospholipid spacing as a potential determinant of intracellular erythrocyte membrane fusion. In particular, we postulate that erythrocyte membranes with tightly-packed phospholipids are fusion-incompetent, whereas those with loosely-packed phospholipids are fusion-competent; and that packing is controlled by the maintenance or loss of the normal asymmetric distribution of phospholipids in the erythrocyte membrane.
