ABSTRACT

The neuroactive kynurenine pathway metabolites quinolinic acid (QUIN) and kynurenic acid (KYNA) have been speculatively linked to the pathogenesis of several neuro-psychiatric diseases (Schwarcz, 1992;Heyes, 1993;Reinhard et al., 1994). The idea that QUIN or KYNA may be causally involved in brain disorders derived originally from the discovery that intracerebral application of QUIN results in excitotoxic neurodegeneration which resembles the lesions seen in Huntington’s disease, temporal lobe epilepsy and cerebral ischemia, that KYNA can serve as a potent neuroprotectant against excitotoxic injury, and that both compounds are normal constituents of the mammalian brain. Subsequently, individual enzymes of the kynurenine pathway were shown to be present in the brain, and abnormal enzyme activities and metabolite levels were found in brain tissue in a number of human brain diseases (Schwarcz and Du, 1991; Stone, 1993).
