ABSTRACT

The octapeptide hormone of the renin angiotensin system, angiotensin II (AII), regulates a wide variety of physiological responses including salt and water balance, blood pressure, and vascular tone. All is produced both systemically and locally in the vessel wall by the actions of renin, which converts angiotensinogen into AI, and angiotensinconverting enzyme (ACE), which cleaves AI to form AII. In addition to its vasoconstrictor activity, AII has recently been shown to stimulate vascular smooth muscle cell (VSMC) growth, increase the expression of inflammatory mediators such as phospholipase A2 and NADH oxidase, and activate gene transcription of proto-oncogenes such as c-fos, as well as genes activated by the signal transducers and activators of transcription (STAT) pathway. These data suggest that AII plays an important role in various cardiovascular diseases associated with VSMC growth and vessel wall inflammation such as hypertension, restenosis following balloon angioplasty, and atherosclerosis.
