ABSTRACT

Eine spezifische Blockade von Hsp90 mittels der Geldanamycin-Derivate 17AAG und 17DMAG führt zu einer signifikanten Inhibition von pro-angiogenetischen Signalkaskaden in humanen Magenkarzinomzellen, die insbesondere unmittelbar das EGFR/Her-2 Rezeptorsystem als auch die Transkriptionsfaktoren STAT3 und HIF-1α betreffen. Daruber hinaus vermag 17DMAG das Tumorwachstum und die Angiogenese des Magenkarzinoms in einem experimentellen Modell zu inhibieren. Hsp90 Inhibitoren stellen demnach einen interessanten Ansatzpunkt für die Entwicklung von molekularen antiangiogenetischen/antineoplastischen Therapiekonzepten zur Behandlung des Magenkarzinoms dar.
