ABSTRACT

A process for production of a target recombinant polypeptide in a prokaryotic cell is provided. The process comprises expressing a vector comprising a first expression cassette comprising a polynucleotide encoding the target polypeptide operably linked to an inducible promoter and comprising a ribosome binding site; and a second expression cassette comprising a polynucleotide encoding an sRNA which targets the ribosome binding region of mRNA which corresponds to the ribosome binding site of the first expression cassette, operably linked to a promoter and wherein the sRNA comprises a sequence of bases complementary to at least a part of the ribosome binding region of the mRNA, which sequence comprises at least 8 consecutive bases in which no more than 15% of the bases exhibit non-Watson-Crick nucleotide base pairing to the ribosome binding region of the mRNA.
