ABSTRACT

In vitro method of separating, detecting or enriching different DNA species present in a sample, on the basis of non-methylated CpG-dinucleotides occurring with varying frequency, present within each DNA species, comprises: isolating DNA containing different DNA species from the sample and preparing an aqueous solution of the isolated DNA; contacting the DNA solution with at least one protein binding to the non-methylated CpG dinucleotides immobilized on a carrier material; and leaching the non-specifically bound DNA and methylated DNA. In vitro method of separating, detecting or enriching different DNA species simultaneously present in a sample, on the basis of non-methylated CpG-dinucleotides occurring with varying frequency, present within each DNA species, comprises: isolating DNA containing different DNA species from the sample and preparing an aqueous solution of the isolated DNA; contacting the DNA solution with at least one protein binding to the non-methylated CpG dinucleotides immobilized on a carrier material; leaching the non-specifically bound DNA and methylated DNA; eluting the carrier material with at least one eluant having different concentrations; correlating the certain regions of concentration of the used eluent with certain CpG-dinucleotide frequencies in the present DNA species so that different fractions of DNA species with different frequencies of CpG dinucleotides are obtained during the elution; and originating the different DNA species from different bacteria and/or fungi.
