ABSTRACT

A method for determining the sequence of a dna, comprising the carrying out the following steps for each radical of the dna to be sequenced: (a) contacting the dna with a dna - polymerase in the presence of (i) a primer and (ii) four nucleotide analogues under conditions which allow it to the dna - polymerase, dna - synthesis to catalyze, wherein (1) the nucleotide analogues of an analogue of dgtp, an analog of dctp, is an analogue of dttp or dutp as well as an analogue of datp, (2) of each nucleotide analog comprises: (i) a base selected from the group consisting of adenine, guanine, cytosine, thymine or uracil and analogs thereof, (ii) a deoxy ribose, (iii) is a radical, which in detachable manner to the 3 '- oxygen of the deoxy ribose is bonded, and (iv) a specific marking which, in a detachable manner to the base is bound, so that comprises one, which is complementary to the sequenced radical, by means of the dna - polymerase in the dna is integrated, and (3) of each..
